Instead of just using the Spotify web player in Chrome, download the actual spotify app, from their website. Don't get it from the windows store as that may cause accessibility issues. The spotify app has its own volume control which is independent of the rest of your system, you can adjust it by pressing CTRL+Down to decrease or CTRL+Up to increase it.
The other thing you can do is use the audio ducking feature of JAWS, which works like the audio ducking feature on iPhone or Android so it'll lower the volume of all other sounds while JAWS speaks. To toggle it on and off I think the keystroke was JAWS+Space, then D. You can also adjust this in the settings center.

You can also use the Windows volume mixer to lower the Google Chrome volume. It allows you to set the relative volume for every application on your system.
